Structural Diversity using Hyp “Customizable Units” : Proof?of?Concept Synthesis of Sansalvamide?Related Antitumoral Peptides

The potential of “customizable units” to generate structural diversity for biological screenings is highlighted in this proof-of-concept synthesis new peptides related the potent antitumoral Sansalvamide A. Using L-4-hydroxyproline (Hyp) as a customizable unit linear parent peptide, an improved procedure selective peptide modification was developed. A divergent Hyp scission-reductive amination process carried out, affording five with cationic residues, and notably, N-alkyl moiety that affected conformation peptide. After two steps (saponification macrocyclization), sixteen differently N1-substituted cyclic were obtained. For first time, activity compounds compared. Not only some analogs but also scarcely studied residues active against MCF7 breast cancer line. Thus, generated from units can be valuable drug discovery.
